Competences:
IEB is a public research organization that belongs to the Academy of Sciences of the Czech Republic. The mandate of IEB is to conduct basic and applied research in the area of plant genetics, physiology and biotechnology.IEB has a unique expertise in flow cytogenetics in plants. They developed novel methods for preparation of chromosome suspensions suitable for flow cytometry and for chromosome sorting and showed that flow-sorted plant chromosomes were suitable for physical gene mapping, construction of chromosome-specific DNA libraries, and targeted isolation of molecular markers A collaborative effort with INRA (France) lead to the creation of a subgenomic BAC library specific for wheat chromosomes 1D, 4D and 6D, and BAC library specific for chromosome 3B of wheat - the first ever chromosome-specific BAC library constructed in plants. Recently, IEB constructed BAC library specific for the short arm of wheat chromosome 1B (1BS) - the first ever chromosome arm-specific BAC library in plants, and for the short arms of wheat chromosome 3A (3AS) and rye chromosome 1R (1RS).
Role:
IEB will isolate wheat chromosomes and chromosome arms 1AS, 1BL, 1BS, 1BL, 3B, 3DS and 3DL by flow sorting and prepare high molecular weight DNA, which will be used to construct subgenomic BAC libraries. The libraries will be ordered in 384 plates and provided to the consortium Beneficiaries. IEB will also construct BAC contig map from chromosome 3DS and anchor the map to the genetic map using available molecular markers and markers developed during the project execution.
Staff members’ profile:
Dr. Jaroslav Dolezel holds a PhD in genetics and is senior research scientist at IEB and associate professor at Palacky University (Olomouc, CZ). He pioneered the development of novel methods of molecular cytogenetics and cytometry to study plant genome structure and evolution. He authored and/or co-authored over 100 papers in peer-reviewed journals and chapters in scientific books. Since 1997 he is the Head of the Olomouc Research Centre of IEB.
Dr. Hana Simkova holds a PhD in genetics and is a senior research scientist at IEB and reader Palacky University. She has a broad experience in plant molecular biology. She pioneered the development of novel methods of chromosome-based genomics in plants, including the construction of BAC libraries from flow-sorted chromosomes. She authored and/or co-authored 11 papers in peer-reviewed journals and 3 chapters in scientific books.
