The construction of anchored physical maps of group 1 and 3 chromosomes in wheat and barley will build on resources and infrastructure developed in WP1 as well as by the associated collaborative projects for barley. WP2 will provide the basis for all consecutive activities planned in the project such as map based cloning (WP3) and molecular markers development for molecular breeding (WP4). The resources and knowledge generated in WP2 will stimulate the improvement of bioinformatics tools and an integrative database in WP5.
In the first phase of the project, all marker resources available for Triticeae will be assessed to define an exhaustive set of group 1 and 3 chromosome markers. Additional markers will be developed in WP1 or through concomitant international collaborative activities. Markers will be used for anchoring BAC contigs (assembling overlapping sequenced fragments) to the genetic map as well as for map-based cloning of genes/traits (WP3) and molecular breeding (WP4). Molecular markers genetically linked to the genes of interest and QTL will be identified. These markers located as close as possible to searched genes will be analyzed in the laboratory in large numbers by using high-throughput genotyping platforms. Thus, when researchers from WP4 will make crossings, the presence of a marker will allow them to quickly verify that the gene is present in plants mothers and their offspring. Researchers will be able to quickly select among a large number of plants, the most powerful individuals.
During the first year, WP2 permitted to (1) establish a database with all potential markers for group 1 and group 3 chromosomes and (2) initiate the development of high-resolution mapping populations in wheat and barley for meiotic anchoring of the physical maps with these markers.
